ABSTRACT This work aimed at investigating the viability of near infrared spectrometry (NIR), associated with chemometric methods, in order to identify differences at the levels of vigor of naturally and artificially aged soybean seeds. Seeds of six soybean genotypes were analyzed when freshly harvested, after natural aging in storage for eight months, and after artificial aging at the temperature of 41 °C for 96 hours. The seed moisture content, germination potential and vigor were evaluated. Also, there were measured the content of protein, oil and of the fatty acids: palmitic, stearic, oleic, linoleic and linolenic. The NIR spectra were obtained from the freeze-dried and grinded seeds. The natural and artificial aging of the seeds promote deterioration at distinct levels, reflecting in differences in seed vigor. The regions of the electromagnetic spectrum between wavelengths of 1000-1200 nm, 1350-1450 nm, 1800-1900 nm and 2300-2400 nm are important to distinguish the levels of quality of the soybean seeds. The contents of oil and protein have a relationship with the physiological quality of the seeds. Also, the most relevant wavelengths for the classification of seed vigor present a relationship with these compounds. NIR spectroscopy, in combination with chemometric methods, presents potential to be used in the classification of soybean seeds regarding their physiological quality.

ABSTRACT. The use of multispectral images has great potential to assess seed quality and represents a significant technological advance in the search for fast and non-destructive analysis techniques. However, the devices currently available are expensive. Thus, this study aimed to propose a low-cost method for acquisition and processing of multispectral images of soybean seeds and to evaluate their potential for rapid determination of seed physiological potential. The study was conducted in three steps: implementation of the multispectral image acquisition system, development of an algorithm for automatic image processing, and evaluation of the relationship between the data obtained through image analysis and the results of standard tests used to evaluate seed physiological potential. A total of 43 variables were assessed, eight related to seed physiological potential (germination and vigor) and 35 obtained from the analysis of the multispectral images. Of the variables obtained from multispectral images, 21 were related to pixel values in the images in the different bands evaluated (green, red, and infrared) and 14 associated with seed morphometric characteristics. The proposed system is efficient in obtaining multispectral images and the algorithm developed was efficient to extract morphometric characteristics and pixel information from the images. The parameters obtained from the NIR spectrum region showed a good relationship with the physiological potential of soybean seeds.

Resumo Soluções baseadas em análise de imagens são uma tendência crescente para melhorar a qualidade das sementes florestais. Este estudo teve como objetivo associar as informações obtidas por meio do processamento de imagens radiográficas com o potencial fisiológico de sementes de Anadenanthera peregrina. Dez lotes de sementes de diferentes matrizes foram submetidos à análise automatizada de raios X para obtenção de variáveis relacionadas à morfologia da semente e integridade dos tecidos. Em seguida, as sementes foram avaliadas quanto ao potencial fisiológico por meio das variáveis germinação, plântulas normais, índice de velocidade de germinação (IVG), comprimento de plântulas, índice de uniformidade e índice de vigor. Os resultados mostraram que houve relação entre as variáveis obtidas com a análise radiográfica e as do potencial fisiológico das sementes. A maioria dos lotes com baixo potencial fisiológico apresentou menor densidade de tecido, circularidade e maior valor de assimetria positiva. A técnica de raios X pode ser considerada válida para estabelecer uma relação entre a qualidade física e fisiológica das sementes, porém, parece ser mais eficiente para identificar sementes com baixo potencial fisiológico.

Abstract Solutions based on image analysis are a growing trend towards improving the quality of forest seeds. This study aimed to associate the information obtained through the processing of radiographic images with the physiological potential of Anadenanthera peregrina seeds. Ten seed lots from different mother trees were submitted to automated X-ray analysis to obtain variables related to seed morphology and tissue integrity. Then, the seeds were evaluated for their physiological potential through the variables germination, normal seedlings, germination speed index (GSI), seedling length, uniformity index, and vigor index. The results showed that there was a relationship between the variables obtained with the X-ray analysis and those of the physiological potential of the seeds. Most of the lots with low physiological potential had lower tissue density, circularity, and a higher positive skewness value. The X-ray technique can be considered valid to establish a relationship between the physical and physiological quality of seeds, however, it seems to be more efficient to identify seeds with low physiological potential.

Resumo: A tecnologia de aplicação do plasma atmosférico consiste em um método rápido e prático, que apresenta potencial de modificação da superfície tegumentar de sementes com dureza tegumentar. O objetivo desta pesquisa foi avaliar o efeito da aplicação do plasma frio atmosférico em sementes de Pityrocarpa moniliformis, verificando suas implicações sobre a impermeabilidade do tegumento, germinação e vigor. As sementes foram submetidas ao plasma por 1,5; 2,0; 3,0; 4,0 e 5,0 minutos, utilizando-se como controle, sementes sem nenhum tratamento. Após a aplicação, determinou-se a molhabilidade do tegumento, curva de embebição e condutividade elétrica da solução de embebição. As sementes também foram submetidas ao teste de germinação, tendo-se, a partir dos seus resultados determinado a viabilidade, mediana, uniformidade e assimetria da germinação. As sementes submetidas ao plasma por 4,0 e 5,0 minutos apresentaram os menores ângulos de contato aparente, 64 e 61°, respectivamente, caracterizando maior molhabilidade do tegumento, entre os tratamentos testados. Maior germinação foi observada nos tratamentos com aplicação do plasma quando comparado com o controle. A tecnologia de aplicação do plasma atmosférico tem potencial para ser utilizada como acelerador da germinação de sementes de P. moniliformis.

Abstract: Atmospheric plasma application technology consists of a fast and practical method, which has the potential to modify the surface of seeds with integumentary hardness. The aim of this research was to evaluate the effect of applying atmospheric cold plasma on Pityrocarpa moniliformis seeds, assessing its implications on the seed coat impermeability, germination and vigor. Cells were subjected to plasma for 1.5, 2.0, 3.0, 4.0 and 5.0 minutes, using seeds without any treatment as a control. After application, the seed coat wettability, imbibition curve and electrical conductivity of the soaking solution were determined. Seeds were also subjected to the germination test, and its results were used to determine viability, median, uniformity and asymmetry of germination. Seeds subjected to plasma for 5.0 and 4.0 minutes showed the lowest apparent contact angles, 64 and 61°, respectively, characterizing greater wettability of the seed coat among the treatments tested. Higher germination was observed in the treatments with plasma application when compared to the control. Atmospheric plasma application technology has the potential to be used as an accelerator of P. moniliformis seed germination.

Resumo: Recentemente está crescendo a necessidade de tecnologias não invasivas, rápidas e precisas que possam estimar a qualidade das sementes. Entre essas tecnologias, destaca-se a análise de imagens de raios X para avaliação da morfologia interna das sementes. Assim, o objetivo deste estudo foi avaliar a eficiência do software SARS na análise de radiografias digitais de sementes de braquiária, denominado SARS (Sistema de Análise de Radiografias de Sementes) na avaliação da qualidade física das sementes. Cinco lotes comerciais de sementes de Urochloa decumbens cv. Basilisk foram utilizados. As sementes foram produzidas na safra de 2018/2019. As imagens radiográficas das sementes foram analisadas por meio do SARS, por meio das quais foram obtidas características físicas. As sementes foram submetidas ao teste de germinação, no qual foram mensuradas variáveis relacionadas à qualidade fisiológica. Foi possível observar que as sementes com maior potencial de germinação e vigor apresentaram correlações fortes e significativas com algumas das variáveis físicas obtidas pelo SARS. Assim, a alta correlação do comprimento de plântulas e a densidade relativa das sementes é importante para validar o método de análise de imagens radiográficas de sementes. O SARS mostrou-se uma ferramenta eficiente para análise de radiografias digitais de sementes de braquiária, gerando descritores que dão suporte a análise morfométrica e interna das sementes, que podem ser associados à sua qualidade fisiológica. Os parâmetros físicos obtidos pela técnica têm estreita relação com a germinação e o vigor das sementes.

Abstract: Recently is growing the need for non-invasive, fast, and accurate technologies that can predict seed quality. Between these technologies, X-ray image analysis stand out for evaluation of the internal morphology of the seeds. Thus, the aim of the present study was to evaluate the efficiency of a specialized software for analyzing digital radiographs of Urochloa decumbens seeds called SARS (Sistema de Análise de Radiografias de Sementes - Seed Radiograph Analysis System). Five comercial seed lots of U. decumbens cv. Basilisk were used. The seed lots were produced in the 2018/2019 crop season. Radiographic images of the seeds were analyzed in SARS, through which physical characteristics were obtained. The seeds were then subjected to germination test, in which variables related to the physiological quality were evaluated. It was possible to observe that the seeds with greater germination and vigor showed strong and significant correlations with some of the physical variables obtained using SARS. Thus, high correlation of seedling length and relative seed density is important for validating the seed radiographic image analysis method. SARS proved to be an efficient tool for analyzing digital radiographs of U. decumbens seeds. It can generate descriptors which support morphometric and internal analysis of the seeds. Physical parameters obtained by using the technique have close relationship with the germination and vigor of the seeds.

ABSTRACT: Successive cycles of water absorption and loss favor weathering deterioration, one of the main factors that affect the quality of soybean seeds. This study evaluated the physiological, physical, and morpho-anatomical changes in soybean seeds under weathering deterioration at the pre-harvest phase. Six soybean cultivars (BMX Apolo, DM 6563, NS 5959, NA 5909, BMX Potência, and TMG 1175) were produced in a greenhouse and underwent weathering deterioration through a rainfall simulation system, applying 0, 60, 120, and 180 mm of precipitation at pre-harvest phase. Each rainfall level was divided into two applications at an interval of 72 h: 60 mm (30 + 30), 120 mm (60 + 60), and 180 mm (90 + 90). After harvest, the seeds were evaluated for germination, vigor, physical and morpho-anatomical properties. Weathering deterioration induced by simulated rainfall at the pre-harvest phase contributes to the reduction in soybean seed germination and vigor and is conditioned by the soybean genotype. The increase in intensity of simulated rainfall led to a more significant weathering damage in seeds, as evidenced by the X-ray and tetrazolium test. Cultivars DM 6563 and BMX Potência were more susceptible, while NA 5909 was less susceptible to weathering deterioration (especially at the highest level; 120 mm and 180 mm). Anatomical changes caused by weathering deterioration lead to cell compaction and rupture, mainly in the cell layers of the hourglass and parenchyma, forming intracellular spaces. The presence of weathering damage caused a reduction in physiological soybean seed quality.

Resumo: Tecnologias baseadas em radiação eletromagnética, como a técnica de raios X, contribuíram para estabelecer novas e promissoras metodologias para avaliar a qualidade de sementes. Nesse sentido, este estudo teve como objetivo relacionar parâmetros baseados na análise semiautomatizada de radiografias de sementes de crambe à sua qualidade fisiológica. Imagens radiográficas de sementes pertencentes a 10 lotes da cultivar FMS Brilhante foram analisadas de forma semiautomatizada por meio do software ImageJ®. Foram obtidas medidas de características morfométricas e integridade tecidual das sementes, bem como individualmente para o embrião das sementes. Após o teste de raio-X, as sementes foram submetidas ao teste de germinação e crescimento de plântulas. Foi possível visualizar as estruturas internas das sementes nas radiografias. Houve diferenças nos parâmetros físicos obtidos pela análise semiautomática das radiografias entre os lotes de sementes. Além disso, os lotes diferiram quanto à qualidade fisiológica das sementes. Características morfométricas e de integridade tecidual, especialmente para o embrião, apresentaram alta correlação com a qualidade fisiológica. Assim, este trabalho apresenta uma abordagem eficiente para avaliar de forma rápida e não destrutiva a qualidade de sementes de crambe.

Abstract: Technologies based on electromagnetic radiation, such as the X-ray technique, has contributed to the establishment of new and promising methodologies for evaluating seed quality. This study aimed to relate parameters based on semi-automated analysis of radiographs of crambe seeds to their physiological quality. Radiographic images of seeds from 10 seed lots of cultivar FMS Brilhante were semi-automatically analyzed using ImageJ® software. Measurements of morphometric characteristics and tissue integrity were obtained for the seeds, as well as individually for the seed embryo. Following X-ray test, the seeds were subject to germination and seedling growth test. It was possible to visualize the internal structures of the seeds in the radiographs. There were differences in the physical parameters obtained by the semi-automated analysis of the radiographs between the seed lots. Also, the lots differed regarding the physiological quality of the seeds. Morphometric characteristics and tissue integrity, especially for the seed embryo, showed high correlation with the seed physiological quality. Therefore, this work presents an efficient approach to rapid and non-destructively assess the quality of crambe seeds.

Resumo: A inclusão de técnicas de análise de imagens para a avaliação precisa e rápida da qualidade de sementes florestais é altamente desejável. Entre as técnicas de imagem disponíveis, destacam-se o uso de imagens radiográficas digitais de sementes e a análise de imagens de plântulas, ainda pouco utilizadas na determinação da qualidade de sementes florestais. Objetivou-se com este trabalho, avaliar a viabilidade do uso da análise de imagens de sementes e plântulas na avaliação da qualidade física e fisiológica de sementes de Senna siamea. Imagens radiográficas de sementes de cinco lotes foram obtidas e, a partir delas, foram gerados descritores morfológicos e de integridade tecidual. Após a obtenção das imagens das sementes, estas foram submetidas ao teste de germinação e crescimento de plântulas, dos quais se extraíram variáveis relacionadas à qualidade fisiológica. As plântulas geradas foram escaneadas e analisadas por meio do software ImageJ. Os dados foram analisados por meio da análise de variância, correlação e análise de componentes principais. Os resultados mostraram diferenças entre os lotes de sementes quanto a sua qualidade fisiológica e integridade física de tecidos internos. Foram observadas correlações significativas entre as variáveis obtidas com a análise radiográfica e pelos testes de caracterização fisiológica das sementes. O uso das técnicas de análise de radiografias de sementes e de imagens de plântulas permitem acessar informações de integridade física e fisiológica de sementes de S. siamea.

Abstract: The inclusion of image analysis techniques for the accurate and rapid assessment of the quality of forest seeds is highly desirable. The use of digital radiographic images of seeds and the analysis of seedling images, still little used in determining the quality of forest seeds, are among the available imaging techniques. This study aimed to assess the feasibility of using the analysis of images of seeds and seedlings in the assessment of the physical and physiological quality of Senna siamea seeds. Radiographic images of seeds were obtained from five seed lots, allowing generating morphological and tissue integrity descriptors. These seeds were then subjected to germination and seedling growth tests, which allowed obtaining variables related to physiological quality. The generated seedlings were scanned and analyzed using the software ImageJ. The data were analyzed using analysis of variance, correlation, and principal component analysis. The results showed differences between seed lots in terms of physiological quality and physical integrity of internal tissues. Significant correlations were observed between the variables obtained with the radiographic analysis and seed physiological characterization tests. The use of techniques to analyze seed radiographs and seedling images allows access to information on the physical and physiological integrity of S. siamea seeds.

RESUMO A avaliação da qualidade das sementes de espécies florestais nativas é de extrema importância e, neste sentido, há a necessidade de integrar novas tecnologias que possam complementar as análises de qualidade tradicionalmente utilizadas. O objetivo desse estudo foi avaliar a qualidade física e fisiológica de sementes de Piptadenia gonoacantha utilizando técnicas de análise de imagem e relacionar aspectos de integridade tecidual com atributos fisiológicos. Sementes de 10 lotes foram coletadas e submetidas a análises automáticas e visuais de imagens radiográficas, com a determinação de parâmetros relacionados à densidade tecidual e preenchimento das sementes. Após o teste de raios-X, as sementes foram submetidas a análises fisiológicas de germinação e vigor. Foi observado que os lotes que apresentaram maiores valores de densidade tecidual, preenchimento e solidez, apresentaram maior germinação e vigor. Ficou evidente também que os danos presentes nas sementes tiveram relação direta com o seu desempenho fisiológico. Portanto, os resultados mostraram que a técnica de raios X nas sementes, combinada com a análise automatizada das radiografias, são eficientes na avaliação da morfologia interna de sementes de P. gonoacantha. A integridade física dos tecidos das sementes está diretamente relacionada à qualidade fisiológica e permite a classificação de diferentes lotes quanto ao vigor. A análise visual de radiografias digitais é adequada para identificar a má formação em sementes de P. gonoacantha.

ABSTRACT The assessment of the quality of native forest species seeds is extremely important and, in this sense, it is necessary to integrate new technologies that can complement the quality analysis traditionally used. This study aimed to evaluate the physical and physiological quality of Piptadenia gonoacantha seeds using image analysis techniques and to relate aspects of tissue integrity with physiological attributes. Seeds from 10 lots were collected and submitted to automatic and visual analyses of radiographic images, with the determination of parameters related to tissue density and seed filling. After the X-ray test, the seeds were subjected to physiological analyses of germination and vigor. It was observed that the lots that presented higher values of tissue density, filling, and solidity, presented higher germination and vigor. It was also evident that the damage present in the seeds was causally related their physiological condition. Therefore, the results showed that the use of the X-ray technique in seeds combined with the automated analysis of the radiographs is efficient in the evaluation of the internal morphology of P. gonoacantha seeds. The physical integrity of the seed tissues is directly related to the physiological quality and allows the classification of different lots as to vigor. Visual analysis of digital radiographs is adequate to identify malformation in P. gonoacantha seeds.

RESUMO Informações sobre o padrão da germinação e caracterização de sementes e desenvolvimento de plântulas de Melaleuca alternifolia são escassos. Objetivou-se com este trabalho determinar a temperatura ideal para a germinação de sementes M. alternifolia, além de caracterizar sementes e o desenvolvimento pós-seminal da espécie. Foram utilizados três lotes de sementes de M. alternifolia colocadas para germinar a 20, 25, 20-30, 20-35, 30 e 35 ºC, com as avaliações diárias para determinação da taxa de germinação, índice de velocidade de germinação (IVG), velocidade de germinação (VG), tempo médio para germinação de 50 (t50) e 100% (t100). A morfologia interna das sementes foi avaliada por meio de imagens de raios X e a das plântulas por meio de fotografias. As temperaturas alternadas (20-30 e 20-35 ºC) proporcionaram taxas de germinação superiores às demais. O IVG apresentou os menores e maiores valores nas temperaturas constantes de 20 e 30 ºC para todos os lotes, respectivamente. O VG foi menor na temperatura de 20 ºC e maior na temperatura de 35 ºC. Nas temperaturas alternadas (20-30 e 20-35 ºC), t50 foi em torno de 5 e t100 atingiu 16 dias. Como conclusão, as temperaturas alternadas de 20-30 e 20-35 ºC são indicadas para a germinação de M. alternifolia, com a avaliação final podendo ser realizada aos 16 dias após a semeadura. A análise de raios-X permitiu visualizar as estruturas internas das sementes e distinguir sementes cheias e sementes vazias e impurezas. A germinação é epígea e as plântulas apresentam estabilização do desenvolvimento após 30 dias.

ABSTRACT There is little information regarding the germination pattern, seed characterization, and seedling development of Melaleuca alternifolia. This study aimed to determine the ideal temperature for the germination of M. alternifolia seeds, as well as to characterize the seeds and the post-seminal development of the species. Three lots of M. alternifolia seeds were placed to germinate at 20, 25, 20-30, 20-35, 30, and 35 °C, with daily evaluations to determine germination rate, germination speed index (GSI), speed of germination (SG), and mean time for germination of 50% (t50) and 100% (t100) of seeds. The inner morphology of the seeds was evaluated through X-ray images and seedling morphology by photographs. Alternating temperatures (20-30 and 20-35 °C) led to germination rates higher than the other temperatures. The GSI had the lowest values at the constant temperature of 20 °C and the highest values at the constant 30 °C for all the lots. The SG was lowest at a temperature of 20 °C and the highest at the temperature of 35 °C. At alternating temperatures (20-30 and 20-35 °C), t50 was around 5 days and t100 reached 16 days. In conclusion, the alternating temperatures of 20-30 °C and 20-35 °C are recommended for germination of M. alternifolia, and final evaluation can be performed at 16 days after sowing. The inner structures of seeds could be visualized by X-ray analysis, and full seeds could be distinguished from empty seeds and impurities. Seeds exhibit epigeal germination and seedling development is stabilized after 30 days.

Resumo: Métodos não-destrutivos e de alto desempenho têm sido desenvolvidos para avaliação da qualidade de sementes. O objetivo deste estudo foi relacionar parâmetros obtidos a partir da análise gratuita e automatizada de radiografias digitais de sementes de melão híbrido com o seu potencial fisiológico. Foram utilizadas amostras de sementes comerciais de três cultivares híbridas de melão (Cucumis melo L.), cada uma representada por três lotes. Foram obtidas imagens radiográficas das sementes, das quais foram geradas determinações de área, perímetro, circularidade, densidade relativa, densidade integrada e preenchimento da cavidade interna de sementes, por meio de uma macro (PhenoxXray) desenvolvida para o software ImageJ®. Após o teste de raios-X, as sementes foram submetidas ao teste de germinação, a partir do qual foram obtidas variáveis relacionadas à qualidade fisiológica. Observou-se variabilidade entre lotes para as características físicas e fisiológicas. Os resultados demonstraram que o uso da macro PhenoXray permite a fenotipagem em larga escala das radiografias de sementes de maneira simples, rápida, consistente e totalmente gratuita. A metodologia é eficiente na obtenção de dados morfométricos e de integridade tecidual em sementes de melão, e os parâmetros gerados apresentam estreita relação com atributos fisiológicos da qualidade das sementes.

Abstract: Non-destructive and high throughput methods have been developed for seed quality evaluation. The aim of this study was to relate parameters obtained from the free and automated analysis of digital radiographs of hybrid melons’ seeds to their seeds’ physiological potential. Seeds of three hybrid melon (Cucumis melo L.) cultivars from commercial lot samples were used. Radiographic images of the seeds were obtained, from which area, perimeter, circularity, relative density, integrated density and seed filling measurements were generated by means of a macro (PhenoXray) developed for ImageJ® software. After the X-ray test, seed samples were submitted to the germination test, from which variables related to the physiological quality of the seeds were obtained. Variability between lots was observed for both physical and physiological characteristics. Results showed that the use of the PhenoXray macro allows large-scale phenotyping of seed radiographs in a simple, fast, consistent and completely free way. The methodology is efficient in obtaining morphometric and tissue integrity data of melon seeds and the generated parameters are closely related to physiological attributes of seed quality.

RESUMO Novas e refinadas tecnologias, baseadas em procedimentos computadorizados, criam potencial para automatização de avaliações laboratoriais, alternativa para a redução de custos e aumento da capacidade de tomada de decisão quanto ao destino de lotes de sementes. O presente trabalho teve como objetivo avaliar a eficiência do software de processamento digital de imagens de plântulas, o SAPL®, na caraterização do potencial fisiológico de sementes de milho, em comparação com informações fornecidas por testes de vigor recomendados para essa espécie. Dez lotes de sementes de milho foram submetidos às avaliações de germinação, primeira contagem de germinação, índice de velocidade de germinação, emergência de plântulas, teste de frio e condutividade elétrica. Os resultados desses testes foram comparados com os dados obtidos com a análise de imagem, que avaliou o desenvolvimento de plântulas aos três e cinco dias, contados a partir do início do teste de germinação, mediante a obtenção de valores de comprimento de plântulas, índice de crescimento, uniformidade de desenvolvimento, índice de vigor e índice de vigor corrigido. O SAPL® é uma alternativa consistente e promissora para avaliação do potencial fisiológico de sementes de milho, tendo apresentado alta correlação de suas variáveis com os testes tradicionais utilizados. A avaliação pode ser realizada no terceiro e quinto dia após a instalação do teste de germinação.

ABSTRACT New and refined technologies based on computerized procedures create the potential for automating laboratory evaluations, constituting an alternative to reduce costs and increase the decision-making capacity regarding the destination of seed lots. The objective of this study was to evaluate the efficiency of the Sistema de Análise de Plântulas (SAPL ®) free digital image processing software of seedlings in characterizing the physiological potential of maize seeds in comparison with information provided by vigor tests recommended for this species. Ten lots of maize seeds were submitted to germination, first germination count, germination speed index, seedling emergence, cold test and electrical conductivity. The results of these tests were compared with the data obtained from the image analysis, which evaluated the development of seedlings at three and five days counted from the beginning of the germination test, by obtaining values of seedling length, growth, development uniformity, vigor index and corrected strength index. SAPL ® is a consistent and promising alternative for evaluating the physiological potential of maize seeds, presenting a high correlation of its variables with the traditional tests used. The evaluation can be performed on the third and fifth day after the germination test is installed.

Resumo: O teste de tetrazólio tem sido recomendado para a avaliação da viabilidade de sementes que apresentam germinação lenta e desuniforme, especialmente espécies agroflorestais. O objetivo do trabalho foi propor uma metodologia para avaliação da viabilidade das sementes de J. curcas L. pelo teste de tetrazólio e gerar recursos visuais que auxiliem na interpretação do teste. Nesta pesquisa foram realizados dois ensaios. No primeiro ensaio, foram avaliados métodos de preparo e coloração das sementes. As metodologias mais promissoras foram utilizadas na condução do segundo ensaio, que consistiu na avaliação de diferentes concentrações (0,10 e 0,25%) e tempos (2 e 3 h) de imersão das sementes na solução de tetrazólio, utilizando-se cinco lotes. As sementes foram analisadas individualmente e classificadas como viáveis ou não viáveis. Paralelamente, foram conduzidos testes de germinação, primeira contagem, emergência e índice de velocidade de emergência de plântulas. O teste de tetrazólio é eficiente para a avaliação da viabilidade de sementes de J. curcas L. fornecendo resultados correlacionados com a germinação e a emergência de plântulas. Sementes pré-embebidas devem ter o tegumento removido e, em seguida, seccionadas longitudinal e medianamente no sentido do comprimento para imersão em solução de cloreto 2,3,5-trifenil tetrazólio a 0,10% por três horas a 35 ºC.

Abstract: The tetrazolium test has been recommended for evaluating the viability of seeds with slow and ununiform germination, especially agroforestry species. The aim of this study was to propose a methodology to evaluate the viability of J. curcas L. seeds using the tetrazolium test and to generate visual aids to support the interpretation of the test. In the first trial, methods of seed preparation and staining were evaluated. The most promising methodologies were used to conduct the second test, which consisted of the evaluation of different concentrations (0.10 and 0.25%) and times (2 and 3 h) of immersion of the seeds in the tetrazolium solution, using five lots. The seeds were analyzed individually and classified as viable or non-viable. At the same time, germination, first count, emergence and seedling emergence speed index tests were conducted. The tetrazolium test is efficient for evaluating the viability of seeds of J. curcas L. providing results correlated with germination and seedling emergence. Pre-soaked seeds should have their integument removed and then sectioned longitudinally and medially lengthwise for immersion in 0.10% solution of 2,3,5-triphenyl chloride tetrazolium for three hours at 35 °C.

Resumo: A análise automatizada do vigor de sementes tem se destacado por permitir maior precisão, padronização, objetividade e velocidade na avaliação do potencial fisiológico de lotes de sementes. O presente trabalho teve como objetivo avaliar a eficiência do sistema Vigor-S® na determinação do potencial fisiológico de sementes de feijão comum, em comparação às informações fornecidas por testes de vigor recomendados para essa espécie. Foram utilizados quatro genótipos de feijão comum, cada um representado por quatro lotes. A caracterização do potencial fisiológico dos lotes foi realizada por meio dos testes de germinação, primeira contagem de germinação, emergência de plântulas, envelhecimento acelerado e condutividade elétrica. Os resultados desses testes foram comparados com os dados obtidos com o uso da técnica de análise de imagem, por meio do software Vigor-S®, que foi utilizado para avaliar o crescimento das plântulas aos dois, três e quatro dias após o início do teste de germinação, mediante a obtenção de valores de comprimento da parte aérea, raiz primária, plântula, e os índices de crescimento, uniformidade e vigor. A análise computadorizada de imagens de plântulas utilizando o software Vigor-S® é eficiente para a avaliar o potencial fisiológico de sementes de feijão, de forma similar às avaliações tradicionalmente utilizadas para esta finalidade.

Abstract: Automated analysis of seed vigor stands out by allowing greater accuracy, standardization, objectivity, and speed in evaluation of the physiological potential of seed lots. The objective of this study was to evaluate the efficiency of the Vigor-S® system in assessing the physiological quality of common bean seeds compared to the information provided by the traditional vigor tests recommended for this species. Four genotypes of common bean were used, each one represented by four seed lots. Characterization of the physiological potential of the lots was carried out by the following tests: germination, first count of germination, seedling emergence, accelerated aging, and electrical conductivity. The results of these tests were compared with the data obtained from the image analysis technique, specifically the Vigor-S® system, which was used to evaluate seedling growth at two, three, and four days after the beginning of the germination test. Shoot length, primary root length, and seedling length were measured, as well as the growth index, uniformity index, and vigor index were calculated. Computerized analysis of seedling images using the Vigor-S® software is a reliable alternative for evaluation the physiological potential of bean seeds, and it produces information similar to evaluations traditionally used for that purpose.

Resumo: A necessidade de otimizar a avaliação da qualidade de sementes utilizando novos recursos computacionais mais acessíveis e modernos levou ao surgimento de novas ferramentas. Neste artigo, introduzimos o SeedCalc, um novo pacote do software R desenvolvido para processar dados de germinação e comprimento de plântulas. As funções incluídas no SeedCalc permitem um processamento de dados rápido e eficiente, oferecendo maior confiabilidade às variáveis geradas e facilitando a própria análise estatística, uma vez que os dados já são processados com a estrutura apropriada para serem analisados estatisticamente no software R. Abstract: The need to optimize seed quality assessment using new, more accessible, and modern computational resources has led to the emergence of new tools. In this paper, we introduce SeedCalc, a new R software package developed to process germination and seedling length data. The functions included in SeedCalc allow fast and efficient data processing, offering greater reliability to the variables generated and facilitating statistical analysis itself since the data are already processed with the appropriate structure to be statistically analyzed in the R software.
